This is a combination of three oral Janus kinase (JAK) inhibitors: abrocitinib, baricitinib, and upadacitinib. Each drug selectively inhibits different members of the JAK family, which are intracellular enzymes involved in cytokine signaling via the JAK/STAT pathway. This pathway plays a central role in immune modulation and inflammation. Abrocitinib is primarily a selective JAK1 inhibitor; baricitinib inhibits both JAK1 and JAK2; upadacitinib is also more selective for JAK1 but can inhibit other isoforms at higher concentrations. All three drugs are approved or in advanced clinical development for moderate-to-severe atopic dermatitis and have demonstrated efficacy as monotherapy or with topical corticosteroids[1][2][8]. They are also used individually for other inflammatory diseases such as rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis, Crohn's disease, and ulcerative colitis[6][9]. There is no evidence that this specific triple combination (abrocitinib + baricitinib + upadacitinib) has been developed or studied as a single pharmaceutical product.
