Tyrosine-protein kinase JAK3 (Janus kinase 3, JAK-3) is a non-receptor tyrosine kinase primarily involved in signal transduction for cytokine receptors that share the common gamma chain (γc), such as those for interleukins IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21. It plays a crucial role in hematopoiesis—especially T-cell development—and mediates essential signaling events in both innate and adaptive immunity. Mutations can lead to SCID or leukemia. Selective inhibitors targeting Cys909 are used therapeutically.
