AC-201 is a novel, orally available small molecule that acts as an allosteric inhibitor of tyrosine kinase 2 (TYK2) and Janus kinase 1 (JAK1), binding to the JH2 domain to stabilize its self-inhibitory conformation and thereby inhibit TYK2/JAK1 activity. This mechanism disrupts key cytokine signaling pathways, including those involved in the IL-12/23 and type I interferon pathways, which are implicated in autoimmune diseases such as psoriasis and systemic lupus erythematosus. AC-201 has also demonstrated inhibition of caspase-1 and interleukin-1β (IL-1β) production/activity, with additional effects on down-regulating IL-1β receptors. The drug is being developed primarily for immune-mediated conditions like moderate to severe plaque psoriasis, uveitis, systemic lupus erythematosus, and gout. Clinical trials have shown that AC-201 is safe and well-tolerated in healthy volunteers[7][8][3][4][5].
