Acalabrutinib + bendamustine + rituximab is a three-drug combination regimen approved for the treatment of adult patients with previously untreated mantle cell lymphoma (MCL) who are ineligible for autologous hematopoietic stem cell transplantation. Acalabrutinib is a selective Bruton tyrosine kinase (BTK) inhibitor that blocks B-cell receptor signaling and proliferation. Bendamustine is an alkylating agent that causes DNA crosslinking and apoptosis in malignant cells. Rituximab is a monoclonal antibody targeting CD20 on B lymphocytes leading to their depletion. The combination has demonstrated improved progression-free survival compared to chemoimmunotherapy alone in clinical trials[1][2][3][4][5].
