ACL-513 (also known as CKD-513) is a small molecule histone deacetylase 6 (HDAC6) inhibitor being developed by Chong Kun Dang Pharmaceutical Corp. for the treatment of degenerative neurological diseases. The drug works by selectively inhibiting HDAC6, an enzyme that can disrupt material transport within axons by reducing microtubule stability. By restoring microtubule stability and axonal transport, ACL-513 aims to improve nerve cell function directly. Preclinical studies have shown that the compound possesses high selectivity for HDAC6 and excellent brain penetration, overcoming limitations of previous inhibitors in its class. Research presented at the 2025 Society for Neuroscience Annual Meeting demonstrated its ability to restore axonal transport and recover cognitive function in models of Alzheimer's disease and tauopathy. The project is supported by the Korea Drug Development Fund (KDDF).
