Overview

ACTR087 is an autologous engineered T-cell therapy developed by Unum Therapeutics (now Cogent Biosciences) using its proprietary Antibody-Coupled T cell Receptor (ACTR) platform. The technology, originally developed at the National University of Singapore and St. Jude Children’s Research Hospital, involves engineering T-cells to express a high-affinity CD16 (FcγRIIIa) receptor. Unlike traditional CAR-T therapies that target a single predefined antigen, ACTR087 is designed to be a "universal" T-cell that can be directed against various tumor antigens when administered in combination with specific monoclonal antibodies (mAbs). The T-cells bind to the Fc region of the co-administered antibody, triggering T-cell activation and tumor cell lysis. ACTR087 was clinically evaluated in combination with rituximab for CD20+ non-Hodgkin lymphoma and with SEA-BCMA for multiple myeloma. However, development was discontinued following the termination of a strategic collaboration with Seattle Genetics in 2020 and a shift in company focus toward other programs like ACTR707 and BOXR1030.
