AD-104 is a fixed-dose combination (FDC) drug candidate developed by Addpharma, a subsidiary of Yuhan Corporation, for the treatment of gastrointestinal disorders such as gastroesophageal reflux disease (GERD). Classified as an "Improved New Drug" (IMD) in South Korea, AD-104 combines esomeprazole strontium tetrahydrate, a proton pump inhibitor (PPI), with sodium bicarbonate, an antacid. The formulation is designed to provide an immediate-release profile for esomeprazole; the sodium bicarbonate component neutralizes gastric acid, thereby protecting the acid-labile PPI from degradation in the stomach and facilitating rapid absorption. This mechanism is intended to offer a faster onset of action compared to traditional delayed-release, enteric-coated PPI formulations. AD-104 is currently in Phase 1 clinical development.
