Ad-hDCT is a recombinant adenoviral vector vaccine engineered to express human dopachrome tautomerase (hDCT), a melanocyte-specific enzyme and well-characterized melanoma-associated self-antigen. Developed primarily for use in heterologous prime-boost immunotherapy regimens, Ad-hDCT is often employed as a boosting agent following a priming dose with another viral vector, such as vesicular stomatitis virus (VSV-hDCT). This approach is designed to bypass pre-existing anti-vector immunity and rapidly expand the population of antigen-specific effector CD4+ and CD8+ T cells. In preclinical murine melanoma models, this vaccination strategy has demonstrated the ability to overcome peripheral tolerance to self-antigens, providing significant anti-tumor efficacy in both prophylactic and therapeutic settings.
