Overview

ADG152 is a novel bispecific antibody drug candidate developed by Adagene. It functions as a CD20xCD3 T-cell engager (TCE), designed to redirect T cells to target and kill CD20-expressing tumor cells, such as those found in B-cell malignancies. The molecule incorporates Adagene’s proprietary SAFEbody precision masking technology, which conditionally activates the anti-CD3 arm of the bispecific antibody only in the tumor microenvironment. This approach aims to minimize cytokine release syndrome (CRS) and reduce on-target/off-tumor toxicities that are common with traditional TCEs, thereby improving safety while maintaining potent anti-tumor activity. Preclinical data demonstrate strong efficacy with more than 100-fold control of cytokine release and a 2-3-fold prolonged half-life compared to benchmark antibodies[1][2][4][5][6][8]. As of now, ADG152 remains in preclinical or IND-enabling development.
