AJ1-10502 is a second-generation, small molecule Type II Janus kinase 2 (JAK2) inhibitor being developed by Ajax Therapeutics for the treatment of myeloproliferative neoplasms (MPNs), including myelofibrosis. Unlike first-generation Type I inhibitors (e.g., ruxolitinib) that bind to the active conformation of the kinase, AJ1-10502 specifically targets the inactive (Type II) conformation of JAK2. This mechanism provides enhanced selectivity and the potential for disease modification by reducing the mutant cell fraction in the bone marrow and spleen. The compound was discovered using computational chemistry and structure-based drug discovery platforms in collaboration with Schrödinger, Inc.
