Overview

AK148H1 is a novel bispecific antibody-drug conjugate (ADC) developed by Akeso that simultaneously targets Trophoblast cell-surface antigen 2 (Trop2) and Nectin cell adhesion molecule 4 (Nectin4). Both Trop2 and Nectin4 are frequently overexpressed in a variety of solid tumors, including urothelial, breast, and lung cancers, making them ideal targets for precision oncology. By utilizing a bispecific antibody format, AK148H1 is designed to enhance tumor-binding specificity and potentially overcome resistance mechanisms that limit the efficacy of monospecific ADCs. Upon binding to the target antigens on the tumor cell surface, the ADC is internalized, leading to the intracellular release of a cytotoxic payload that induces cell death. AK148H1 is currently in early-stage development for the treatment of advanced solid tumors.
