ALDHlow-TPDC is an experimental cancer vaccine consisting of dendritic cells (DCs) pulsed with the lysate of tumor cells characterized by low aldehyde dehydrogenase (ALDH) activity. In the context of cancer stem cell (CSC) research, ALDH is often used as a marker to identify and isolate CSCs (ALDHhigh). ALDHlow cells represent the non-stem cell population of the tumor. This vaccine was developed as a control or comparator to CSC-based vaccines (such as CSC-TPDC) to evaluate the specific immunogenicity and therapeutic efficacy of targeting cancer stem cells versus the bulk tumor population. Research conducted at the University of Michigan using murine melanoma (D5) and squamous cell cancer (SCC7) models demonstrated that while ALDHlow-TPDC can induce some anti-tumor immunity, it is significantly less effective than vaccines derived from ALDHhigh CSCs in inhibiting tumor growth and prolonging survival.
