Overview

Alemtuzumab + ofatumumab is an investigational combination immunotherapy consisting of two monoclonal antibodies used to treat chronic lymphocytic leukemia (CLL). Alemtuzumab is a humanized monoclonal antibody directed against the CD52 antigen, a glycoprotein expressed on the surface of normal and malignant B and T lymphocytes, NK cells, and monocytes. Ofatumumab is a human monoclonal antibody that targets a specific epitope encompassing the small and large extracellular loops of the CD20 molecule on B cells. The combination works synergistically to deplete malignant B cells through multiple mechanisms, including antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Clinical studies have evaluated this regimen in treatment-naive CLL patients to achieve high rates of complete remission and minimal residual disease (MRD) negativity.
