ALETA-001 is a first-in-class biologic CAR T-cell engager (CTE) protein developed to enhance the effectiveness of CD19-directed CAR T-cell therapies in patients with B-cell malignancies who have relapsed or are refractory after prior CD19-targeted CAR T therapy. The drug works by binding with high affinity to the CD20 antigen on cancer cells and displaying the CD19 target protein, thereby increasing CD19 antigen density and restoring lost or reduced CD19 expression. This mechanism allows previously administered anti-CD19 CAR T-cells to better recognize and kill cancer cells, overcoming a common resistance mechanism where tumor cells lose or downregulate CD19. ALETA-001 is administered intravenously as a single agent every two weeks in clinical trials[2][4][5][6][7].
