Allo-QuadCAR01 is an allogeneic, off-the-shelf chimeric antigen receptor (CAR) T-cell therapy developed by Avencell Therapeutics for the treatment of relapsed or refractory B-cell malignancies, including various forms of non-Hodgkin lymphoma (NHL) and chronic lymphocytic leukemia (CLL). Utilizing the proprietary QuadCAR platform, the therapy features a single CAR construct that simultaneously targets four distinct B-cell surface antigens: CD19, CD20, CD22, and CD79b. This multi-targeting approach is specifically designed to mitigate the risk of antigen escape, a primary mechanism of resistance and relapse in single-target CAR-T therapies. The product is derived from healthy donor T-cells and engineered using CRISPR/Cas9 technology to knock out the T-cell receptor (TRAC) and HLA class I (B2M) molecules, thereby reducing the potential for graft-versus-host disease (GvHD) and host-mediated rejection. This allogeneic design allows for immediate patient access to treatment without the manufacturing delays associated with autologous CAR-T products.
