Overview

This combination therapy consists of an infusion of donor-derived (allogeneic) natural killer (NK) cells, the anti-CD20 monoclonal antibody rituximab, and the cytokine recombinant human granulocyte-macrophage colony-stimulating factor (rhu-GMCSF, sargramostim). Developed at the M.D. Anderson Cancer Center, the regimen is designed to treat patients with recurrent B-cell malignancies, such as chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma (NHL), following a non-myeloablative allogeneic stem cell transplant. The mechanism involves rituximab-mediated opsonization of malignant B-cells, which are then targeted for destruction by the infused NK cells via antibody-dependent cellular cytotoxicity (ADCC). Sargramostim is administered to enhance the activity and recruitment of the immune effector cells, potentially boosting the ADCC effect and overall anti-tumor response.
