AMG 410

AMG 410 is an investigational small molecule, orally administered, macrocyclic, reversible pan-KRAS inhibitor targeting both the GDP-bound (OFF) and GTP-bound (ON) forms of mutant KRAS proteins. The drug is designed to inhibit a wide array of KRAS mutations and amplifications—excluding HRAS and NRAS—making it a potential precision therapy for tumors driven by KRAS alterations. It has demonstrated significant efficacy in preclinical xenograft models, both as monotherapy and in combination with EGFR and PD-1 targeted therapies. Primary clinical evaluation is in advanced or metastatic solid tumors harboring KRAS mutations, with a particular focus on non-small cell lung cancer and colorectal cancer.
