Amphotericin B plus fluconazole is a combination antifungal therapy that combines two distinct mechanisms of action to treat serious fungal infections. Amphotericin B binds to ergosterol in fungal cell membranes creating pores that lead to cell death, while fluconazole inhibits ergosterol synthesis by blocking 14α-demethylase. This combination has shown efficacy in treating cryptococcal meningitis and other systemic fungal infections, though the interaction between these drugs can be concentration-dependent, ranging from synergistic to antagonistic depending on strain and dosage.
