Overview

Anakinra + teriflunomide is a combination of two immunomodulatory drugs, each with distinct mechanisms of action. Anakinra is a recombinant form of the human interleukin-1 receptor antagonist (IL-1Ra) that competitively inhibits the binding of interleukin-1 (IL-1) to its receptor, thereby reducing inflammation and immune-mediated tissue damage. It is primarily used in rheumatoid arthritis and certain autoinflammatory syndromes[3][9]. Teriflunomide is a small molecule pyrimidine synthesis inhibitor that selectively and reversibly inhibits dihydro-orotate dehydrogenase, leading to reduced proliferation of activated T and B lymphocytes without causing cell death. It is approved for relapsing forms of multiple sclerosis[1][6]. The combination has not been established as a standard therapy; there are no clinical studies or regulatory approvals specifically for this drug pairing. However, both agents individually modulate immune responses through different pathways.
