Antagomir-miR-378a nanoparticle is an experimental RNA-based therapeutic consisting of an antagomir (a chemically modified antisense oligonucleotide) targeting microRNA-378a (miR-378a), encapsulated within a nanoparticle delivery system. miR-378a is a key regulator of metabolic inflammation and lipoprotein metabolism; its upregulation suppresses the estrogen-related receptor gamma (ERRγ) axis and genes involved in fatty acid β-oxidation (FAO) such as PPARα and CPT1α. By silencing miR-378a, the nanoparticle restores ERRγ signaling, enhances mitochondrial FAO, and improves high-density lipoprotein (HDL) metabolism (via LXRα and ABCA1), thereby reducing atherogenic VLDL secretion and metabolic inflammation. This approach is being investigated as a potential therapeutic strategy for atherosclerosis and related metabolic disorders.
