anti-αvβ3 G4 is a fully human IgG4 monoclonal antibody designed to target integrin αvβ3, a marker of tumor stemness and drug resistance in epithelial cancers. Developed by Alpha Beta Therapeutics and researchers at UC San Diego, it is a re-engineered variant of the IgG1 antibody etaracizumab. Unlike its predecessor, which relied on NK cell-mediated cytotoxicity, anti-αvβ3 G4 is optimized to engage tumor-associated macrophages (TAMs) by activating the CD64 (FcγRI) receptor. This engagement triggers TAM-mediated cytotoxicity through the upregulation of inducible nitric oxide synthase (iNOS), demonstrating superior antitumor activity in lung cancer models where NK cell infiltration is limited.
