Overview

Anti-B7-H4 antibodies are a class of immunotherapeutic agents designed to target B7-H4 (also known as B7S1 or VTCN1), a member of the B7 family of immune checkpoint molecules. B7-H4 is frequently overexpressed on the surface of various solid tumors, including breast, ovarian, and endometrial cancers, as well as on tumor-associated macrophages, where it acts as a potent inhibitor of T-cell-mediated immunity. By binding to B7-H4, these antibodies aim to block its inhibitory signaling, thereby restoring T-cell proliferation, cytokine production (such as IFN-γ), and anti-tumor activity. Several formats are under investigation, including monospecific monoclonal antibodies, antibody-drug conjugates (ADCs), and bispecific T-cell engagers.
