Overview

Anti-B7H3 monoclonal antibodies are a class of immunotherapeutic agents designed to target **B7-H3** (also known as **CD276**), a member of the B7 family of immune checkpoint proteins. B7-H3 is frequently overexpressed in various solid tumors, including neuroblastoma, melanoma, small cell lung cancer, and sarcomas, while showing limited expression in normal tissues, making it an attractive target for cancer therapy. These antibodies typically work by binding to B7-H3 on the tumor cell surface, potentially blocking its inhibitory signals to T cells or mediating tumor cell destruction through antibody-dependent cellular cytotoxicity (ADCC). Several specific candidates, such as enoblituzumab and omburtamab, have been investigated in clinical trials for various oncological indications, and the anti-B7H3 backbone is also utilized in the development of bispecific antibodies and antibody-drug conjugates (ADCs).
