This combination therapy consists of three distinct chimeric antigen receptor (CAR) T-cell products targeting CD19, CD20, and CD22, administered as a sequential infusion or cocktail, in combination with chidamide (tucidinostat), a selective histone deacetylase (HDAC) inhibitor. Developed by the Affiliated Hospital of Xuzhou Medical University, this regimen is designed to treat refractory or relapsed B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL) and various B-cell lymphomas. The rationale for the triple-target approach is to mitigate immune escape caused by antigen loss, a frequent mechanism of resistance in single-target CAR-T therapies. Chidamide is added to the regimen to potentially upregulate target antigen expression on malignant cells and modulate the tumor microenvironment, thereby enhancing the persistence and cytotoxic efficacy of the CAR-T cells.
