Anti-CD19 CAR-T lentiviral vector is a gene therapy tool designed for the generation of CAR-T cells, specifically optimized for *in vivo* applications. It consists of a lentiviral vector (LVV) encoding a chimeric antigen receptor (CAR) that targets the CD19 antigen, a protein highly expressed on B-cell malignancies. The vector is often pseudotyped with specific envelope proteins, such as the cocal-pseudotyped envelope, to enhance its ability to transduce resting T cells. Developed and manufactured through collaborations involving Shenzhen Eureka Biotechnology, Vector Builder, and Cytiva, this platform utilizes stable producer cell lines (SPCLs) in 293T cells. This manufacturing approach aims to provide greater lot-to-lot consistency, scalability, and an improved safety profile by reducing the release of cytokines associated with cytokine release syndrome (CRS) compared to traditional transient transfection methods. The vector construct typically includes a reporter gene, such as EGFP, linked via an IRES element to facilitate monitoring of transduction efficiency.
