Anti-CD20 CAR-NK cells are an investigational cell therapy platform consisting of natural killer (NK) cells genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD20 antigen, which is primarily expressed on B cells. This modality combines the innate cytotoxic capabilities of NK cells with the antigen-specific targeting of a CAR. Research, including studies by the National Institutes of Health (NIH) and MD Anderson Cancer Center, has explored "armoring" these cells with cytokines like interleukin-15 (IL-15) to enhance in vivo persistence and expansion. In autologous rhesus macaque models, these cells have demonstrated the ability to effectively target B cells and traffic into tissues, although the inclusion of IL-15 may not significantly enhance short-term expansion in certain settings. This platform is primarily developed for the treatment of B-cell malignancies such as non-Hodgkin lymphoma and chronic lymphocytic leukemia.
