anti-CD20 panCAR is an investigational in vivo chimeric antigen receptor (CAR) therapy developed by Orna Therapeutics. It utilizes a synthetic circular RNA (oRNA) platform delivered via proprietary lipid nanoparticles (LNPs) to transiently reprogram T cells in vivo to express an anti-CD20 CAR. This approach enables scalable and non-viral delivery of CAR constructs directly into the patient’s T cells without the need for ex vivo cell manipulation or lymphodepletion regimens. Preclinical studies have demonstrated robust and sustained B cell depletion in non-human primates following a single dose, with up to 95% reduction of B cells and sustained effects observed for at least 7 days post-dosing. The primary indications are B cell-mediated diseases, including oncology and autoimmune disorders[5][8].
