Drug intelligence / Profile preview
An anti-CD20xCD40 bispecific antibody is a dual-targeting immunotherapeutic agent designed to simultaneously bind CD20 on B cells and CD40 on antigen-presenting cells (such as dendritic cells, B cells, and monocytes). This bispecific modality aims to achieve deep, multi-lineage immunomodulation by combining CD20-mediated B-cell depletion with the modulation of CD40-mediated cellular and humoral immune pathways. Preclinical candidates, such as DFI201 and DFI205 developed by Diabetes Free, Inc. (DFI, Inc.), have demonstrated potent B-cell depletion, clearance of tissue-resident memory B cells, transient reduction of non-classical monocytes, and depletion of dendritic cell subsets. Additionally, these bispecific antibodies promote the expansion of regulatory T cells (Tregs) and induce T-cell exhaustion without triggering broad effector T-cell activation or cytokine release syndrome. This dual mechanism of action is being investigated for the treatment of systemic autoimmune rheumatic diseases (SARDs) and other autoimmune disorders to achieve a durable immune reset.
