An anti-CD244 monoclonal antibody is a therapeutic biologic designed to target the CD244 (2B4) receptor, a member of the signaling lymphocyte activation molecule (SLAM) family. CD244 is primarily expressed on natural killer (NK) cells and cytotoxic T lymphocytes, where it typically functions as an inhibitory checkpoint receptor. In the tumor microenvironment, chronic antigen exposure leads to the upregulation of CD244, contributing to T-cell exhaustion and impaired anti-tumor immunity. By blocking the interaction between CD244 and its ligand (CD48), these antibodies aim to reinvigorate effector immune cells. Research indicates that CD244 blockade can synergize with other treatments, such as radiotherapy, by reducing the infiltration of myeloid-derived suppressor cells (MDSCs) and promoting a more robust, cytotoxic CD8+ T-cell response.
