**anti-CD38 mAb + anti-CD3xCD20 bsAb** is an investigational combination therapy consisting of a monoclonal antibody targeting CD38 and a bispecific antibody targeting CD3 on T cells and CD20 on B cells. The anti-CD38 mAb, such as daratumumab or isatuximab, binds to CD38 on malignant plasma cells to induce antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and direct apoptosis, primarily for multiple myeloma. The anti-CD3xCD20 bsAb recruits T cells to CD20-positive B cells, enabling T-cell-mediated lysis, effective against B-cell lymphomas and potentially addressing CD20 downregulation resistance. This combination aims to synergistically target both plasma cell and B-cell malignancies, leveraging complementary mechanisms for enhanced antitumor activity in relapsed/refractory settings.
