Overview

Anti-CSPG4 CAR-macrophages are an experimental cell therapy consisting of primary human macrophages engineered to express a chimeric antigen receptor (CAR) targeting chondroitin sulfate proteoglycan 4 (CSPG4). CSPG4 is a surface antigen highly expressed in melanoma and other solid tumors. Unlike CAR T-cells, CAR-macrophages are designed to penetrate physical tumor barriers and promote phagocytosis of cancer cells. Research from the University of Utah indicates that while these cells can efficiently phagocytose melanoma cells, their cytotoxic activity is significantly enhanced when combined with anti-CD47 blocking antibodies, which disrupt the "don't eat me" signaling axis. Preliminary in vivo studies show these cells can delay tumor growth in xenograft models.
