The anti-human CTLA-4×TIGIT bispecific antibody is an investigational immunotherapy designed to selectively target and deplete tumor-infiltrating FOXP3+ regulatory T cells (Tregs). By simultaneously binding to Cytotoxic T-Lymphocyte-Associated Protein 4 (CTLA-4) and T cell Immunoreceptor with Ig and ITIM domains (TIGIT), which are highly co-expressed on effector Tregs within the tumor microenvironment, the antibody facilitates targeted Treg depletion via antibody-dependent cellular cytotoxicity (ADCC). This dual-targeting approach aims to overcome the systemic toxicity associated with broad peripheral Treg depletion observed with conventional anti-CTLA-4 monotherapies. Developed through a collaboration involving Logone Bio-Convergence Research Foundation, BitD Inc., and Seoul National University, the antibody has demonstrated potent antigen blocking and enhanced binding affinity in acidic tumor conditions in preclinical models of non-small cell lung cancer (NSCLC).
