Anti-IL-22 antibodies are biologic agents designed to neutralize the cytokine interleukin-22 (IL-22), a member of the IL-10 family primarily produced by Th17, Th22, and innate lymphoid cells. IL-22 signals through a heterodimeric receptor complex (IL-22R1 and IL-10R2) to activate the STAT3 pathway, playing a critical role in epithelial barrier maintenance and antimicrobial defense. However, dysregulated IL-22 expression is implicated in the pathogenesis of inflammatory skin diseases, such as atopic dermatitis and psoriasis, where it drives epidermal hyperplasia and barrier dysfunction. In oncology, IL-22 is associated with tumor cell proliferation, stemness, and aggressive phenotypes in cancers like colorectal cancer. Therapeutic blockade of IL-22 aims to restore skin homeostasis in inflammatory conditions and inhibit inflammation-induced tumorigenesis. Several companies, including BioAtla and Anthrobody, are developing potent, humanized, or fully human anti-IL-22 antibodies to address these unmet needs, following earlier clinical development of agents like fezakinumab.
