Overview

An anti-IL-4Ra antibody is a monoclonal antibody designed to bind to the alpha subunit of the interleukin-4 receptor (IL-4Ra). This receptor subunit is a critical component of both the Type I receptor complex (activated by IL-4) and the Type II receptor complex (activated by both IL-4 and IL-13). By targeting IL-4Ra, these antibodies provide dual blockade of IL-4 and IL-13 signaling pathways, which are central drivers of Type 2 (Th2) inflammation. This mechanism is highly effective in treating atopic and allergic diseases, as it inhibits the activation of immune cells like eosinophils and mast cells, as well as structural cells like fibroblasts and keratinocytes. The most prominent example of this class is dupilumab, developed by Regeneron and Sanofi, which has demonstrated significant clinical benefit in reducing skin inflammation, epidermal hyperplasia, and itch in patients with atopic dermatitis.
