The anti-IL10RB blocking mAb is a monoclonal antibody designed to target and inhibit the Interleukin 10 receptor subunit beta (IL10RB). IL10RB is a shared signaling subunit for several cytokines in the IL-10 family, including IL-10, IL-22, IL-26, and IFN-lambdas. Research presented by the National Cancer Center Research Institute (Japan) indicates that IL10RB is significantly upregulated during epithelial-mesenchymal transition (EMT), specifically regulated by the transcription factor Snail. This upregulation is linked to increased tumor malignancy, including enhanced cell invasion, adhesion, chemoresistance, and the formation of invadopodia. Furthermore, IL10RB expression is associated with systemic immune dysfunction, characterized by a reduction in T cell and NK cell populations and their cytotoxic activity. In preclinical models, treatment with an anti-IL10RB blocking antibody has demonstrated significant anti-tumor effects and the potential to reverse immune suppression, suggesting it as a novel therapeutic strategy for overcoming tumor escape and treatment resistance in advanced cancers.
