anti-IL1RAP ADC

anti-IL1RAP ADC is an antibody-drug conjugate (ADC) being developed by Cantargia AB in collaboration with AbbVie. It targets Interleukin-1 receptor accessory protein (IL1RAP), a protein overexpressed in various malignancies—including acute myeloid leukemia (AML), pancreatic, lung, renal, urothelial, and ovarian cancers—as well as within the tumor microenvironment on cancer-associated fibroblasts and suppressive immune cells. The ADC consists of a monoclonal antibody conjugated to a potent maytansinoid payload via a cleavable linker. It is designed to selectively deliver the cytotoxic agent to IL1RAP-expressing cells, exerting anti-tumor activity through direct cell killing and modulation of the tumor microenvironment. Preclinical results presented at AACR 2025 demonstrated significant dose-dependent anti-tumor activity and tumor regression in multiple in vivo models, including melanoma and pancreatic cancer models.
