Anti-IL6R refers to a class of monoclonal antibodies designed to bind to and inhibit the interleukin-6 receptor (IL-6R). By blocking the binding of the pro-inflammatory cytokine IL-6 to its receptor, these agents inhibit the downstream signaling pathways that drive systemic inflammation and the acute phase response. This mechanism is particularly relevant in conditions characterized by hyperinflammation or "cytokine storms," such as rheumatoid arthritis, giant cell arteritis, and severe COVID-19. Prominent examples of drugs in this category include tocilizumab and sarilumab. Clinical research, such as studies involving the Sepsis Response Signature (SRS-1) endotype, has demonstrated that IL-6R blockade can rescue antigen presentation and reverse immune dysregulation in specific patient subsets presenting with febrile respiratory illness.
