Overview

Anti-ITGB1 antibody refers to a class of monoclonal antibodies designed to target and inhibit integrin beta-1 (ITGB1), also known as CD29. ITGB1 is a key cell surface receptor that mediates cell-extracellular matrix adhesion and downstream signaling pathways involved in cell migration, survival, and proliferation. In H3K27M-mutant diffuse midline gliomas (DMG), ITGB1 expression is associated with higher tumor grade and poor prognosis. Inhibition of ITGB1 has been shown to disrupt tumor cell migration, reduce MYC-driven purine biosynthesis, and decrease the stemness of glioma cells. Preclinical studies, particularly those involving CNS-directed delivery, have demonstrated that anti-ITGB1 antibodies can extend survival in DMG models, especially when used in combination with inhibitors of other integrins (such as ITGB3 and ITGB5) to prevent compensatory signaling.
