Anti-p21Ras single-chain variable fragment (scFv) is an intracellularly acting biologic therapeutic designed to target the p21Ras protein, a product of the RAS oncogenes (H-ras, K-ras, and N-ras) that is frequently mutated or overexpressed in human cancers. The scFv consists of 246 amino acids, comprising variable heavy (VH) and light (VL) chain regions joined by a 15-amino acid flexible polypeptide linker. Because the Ras protein is located on the inner surface of the plasma membrane, the scFv requires specialized delivery mechanisms to enter cells, such as recombinant adenovirus vectors (e.g., KGHV100, KGHV300, KGHV500) or conjugation with cell-penetrating peptides like RGD4C. Preclinical research, primarily conducted by academic institutions associated with the KGH-R1 hybridoma line, has demonstrated its ability to inhibit tumor cell growth in colorectal cancer and other solid tumors by blocking Ras-mediated signaling pathways.
