Overview

This is a combination therapy consisting of an anti-PD-1 monoclonal antibody, chidamide, and rituximab. Anti-PD-1 antibodies are immune checkpoint inhibitors that block the programmed cell death protein 1 (PD-1) pathway, thereby enhancing T-cell mediated immune responses against cancer cells. Chidamide (also known as tucidinostat) is a selective histone deacetylase (HDAC) inhibitor that targets HDAC1, HDAC2, HDAC3, and HDAC10; it modulates gene expression and can sensitize tumor cells to other therapies by increasing CD20 expression on B-cells. Rituximab is a chimeric monoclonal antibody targeting CD20 on B lymphocytes and is widely used in the treatment of B-cell malignancies such as diffuse large B-cell lymphoma (DLBCL). The combination aims to leverage immune activation via PD-1 blockade with enhanced tumor cell susceptibility from epigenetic modulation by chidamide and direct cytotoxicity from rituximab[4][5][6][7].
