Overview

Anti-PD1 monotherapy refers to the therapeutic use of a single-agent monoclonal antibody that targets the Programmed Cell Death Protein 1 (PD-1) receptor. PD-1 is a key immune checkpoint protein expressed on the surface of T cells; when it binds to its ligands, PD-L1 or PD-L2, it transmits an inhibitory signal that reduces T-cell activation and proliferation. By blocking this interaction, anti-PD1 antibodies effectively 'release the brakes' on the immune system, enhancing the ability of T cells to recognize and eliminate cancer cells. This treatment modality is a cornerstone of modern oncology and is utilized across a broad spectrum of malignancies, including melanoma, non-small cell lung cancer, and renal cell carcinoma. Recent clinical reports also highlight its efficacy in specific subsets of pediatric brain tumors, such as replication repair deficient (RRD) medulloblastoma.
