Overview

Anti-PLXNB2 monoclonal antibodies are therapeutic agents designed to target Plexin-B2 (PLXNB2), a transmembrane protein identified as a key driver of circulating tumor cell (CTC) clusters and metastatic progression. By neutralizing PLXNB2, these antibodies disrupt its interactions with ligands such as SEMA4C on tumor cells and SEMA4A on myeloid cells, thereby inhibiting both homotypic tumor-tumor and heterotypic tumor-myeloid adhesion. This mechanism aims to prevent the formation of CTC clusters, which are significantly more efficient at mediating metastasis than single CTCs. Research led by Northwestern University has utilized AI-guided structural modeling and language models to optimize these antibodies for enhanced affinity and neutralization, specifically targeting breast cancer metastasis.
