Overview
This is a combination prophylactic regimen consisting of low-dose anti-thymocyte globulin (ATG), post-transplant cyclophosphamide (PTCy), and ivarmacitinib (SHR0302). It is being evaluated for the prevention of acute graft-versus-host disease (aGVHD) in patients undergoing haploidentical peripheral blood stem cell transplantation, particularly those with high-risk donors such as parous females. Ivarmacitinib is a selective, oral Janus kinase 1 (JAK1) inhibitor developed by Jiangsu Hengrui Medicine. The regimen aims to leverage the T-cell depleting effects of ATG and PTCy alongside the targeted anti-inflammatory signaling inhibition of ivarmacitinib to reduce the incidence of grade II-IV aGVHD while maintaining graft-versus-leukemia effects and preserving hematopoiesis.
