Overview

Anti-TRP1 CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat melanoma by targeting Tyrosinase-related protein 1 (TRP1). TRP1 is a lineage-specific antigen highly expressed in both melanoma cells and normal melanocytes. The CAR construct typically utilizes a single-chain variable fragment (scFv) derived from an anti-TRP1 monoclonal antibody, linked to 4-1BB (CD137) and CD3ζ signaling domains to promote T-cell persistence and activation. Developed by researchers at the Moffitt Cancer Center, this therapy has shown the ability to significantly reduce tumor growth and improve survival in preclinical melanoma models. While targeting TRP1 can lead to vitiligo due to its presence in healthy melanocytes, the condition is non-life-threatening, making TRP1 a viable target for solid tumor immunotherapy.
