AP Interceptors (also known as Apertor Interceptors) are a proprietary class of proximity-inducing bisteric small molecules developed by Apertor Pharmaceuticals. These bifunctional compounds are designed to traverse the cell membrane and recruit members of the intracellular FKBP (FK506-binding protein) family to a specific oncogenic target protein. By inducing a proximity-based neo-interface, AP Interceptors selectively disrupt protein-protein interactions (PPIs) that are critical for the function of driver proteins such as KRAS. This approach is particularly effective for targeting shallow chemical interfaces on proteins that are traditionally considered undruggable by standard small-molecule inhibitors. Preclinical data has demonstrated their ability to durably block the growth of cancer cells harboring KRAS alterations.
