Overview

AR-13154 is a small molecule drug developed by Aerie Pharmaceuticals for ophthalmic indications, particularly retinal diseases. It acts as a multi-targeted kinase inhibitor, specifically inhibiting Rho kinase (ROCK), Janus kinase 2 (JAK2), and platelet-derived growth factor receptor beta (PDGFRβ). These targets are implicated in the pathological processes of retinal neovascularization, vascular dysfunction, fibrosis, and inflammation. Preclinical studies have shown that AR-13154 can significantly reduce choroidal neovascular lesion size in animal models of wet age-related macular degeneration (AMD), with efficacy comparable to or greater than current anti-VEGF therapies such as aflibercept. The drug has also been evaluated for sustained delivery using bioresorbable polymer implants and microparticle technologies to enable long-term intraocular release[1][3][4][6][8][9].
