Drug intelligence / Profile preview
ARC-02 + rituximab is an investigational combination regimen being evaluated for the treatment of B-cell non-Hodgkin lymphoma (NHL), including follicular lymphoma (FL), diffuse large B-cell lymphoma (DLBCL), marginal zone lymphoma (MZL), and mantle cell lymphoma (MCL). ARC-02 is a next-generation antibody-drug conjugate (ADC) targeting CD79b, developed using Araris Biotech's proprietary AraLinQ site-specific conjugation technology and Arastatin linker-payload. It delivers the cytotoxic microtubule inhibitor monomethyl auristatin E (MMAE) directly to CD79b-expressing B-cells. Rituximab is an approved chimeric monoclonal antibody targeting CD20, which mediates B-cell depletion through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The combination is designed to synergistically target B-cell malignancies through dual antigen targeting (CD79b and CD20) and complementary mechanisms of action. The regimen is currently undergoing evaluation in a Phase 1 clinical trial (ABLATE-101; NCT07691606).
