Overview

This combination therapy consists of the PD-L1 inhibitor atezolizumab, the anti-CD20 monoclonal antibody obinutuzumab, and the immunomodulatory agent lenalidomide. It is being investigated for the treatment of relapsed or refractory follicular lymphoma. Atezolizumab works by blocking the interaction between PD-L1 and its receptors, thereby restoring T-cell anti-tumor activity. Obinutuzumab targets CD20 on B-cells to induce direct cell death and antibody-dependent cellular cytotoxicity. Lenalidomide enhances the immune response by modulating the activity of T cells and NK cells and inducing the degradation of specific transcription factors via the cereblon E3 ubiquitin ligase complex. The combination aims to leverage synergistic effects between checkpoint inhibition, B-cell depletion, and immune modulation.
