ATI-1777 (also known as M520102) is a topical "soft" Janus kinase (JAK) inhibitor being developed for the treatment of atopic dermatitis. Originally developed by Aclaris Therapeutics and licensed to Maruho for development in Japan, the compound is designed to selectively inhibit JAK1 and JAK3. As a "soft" drug, ATI-1777 is engineered to be metabolically unstable in the systemic circulation; it remains active at the site of application on the skin but is rapidly degraded into an inactive metabolite once it enters the bloodstream. This design potentially reduces the systemic side effects typically associated with oral JAK inhibitors while maintaining potent local anti-inflammatory activity. It is currently being evaluated in Phase 2 clinical trials as an ointment for patients with moderate-to-severe atopic dermatitis.
