ATI-501

ATI-501 is an oral small molecule inhibitor targeting Janus kinase 1 (JAK1) and Janus kinase 3 (JAK3). It was developed by Aclaris Therapeutics for the treatment of alopecia areata and its more severe forms, alopecia totalis and alopecia universalis. The drug acts by inhibiting JAK1 and JAK3 enzymes involved in the JAK/STAT signaling pathway, which plays a key role in immune-mediated diseases such as alopecia areata. Clinical trials demonstrated that ATI-501 achieved statistically significant improvements over placebo in hair regrowth measures but did not show clear advantages over other oral JAK inhibitors. The drug was generally well-tolerated with no serious adverse events reported; most adverse events were mild or moderate. Development for alopecia indications has been discontinued after Phase 2 trials[1][2][5][8].
